A comparison of in situ vs. ex situ filtration methods on the assessment of dissolved and particulate metals at hydrothermal vents ArchiMer

The objective of this study was to assess the impact of the filtration method (in situ vs. ex situ) on the dissolved/particulate partitioning of 12 elements in hydrothermal samples collected from the Lucky Strike vent field (Mid-Atlantic Ridge; MAR). To do so, dissolved ( <0.45 mu m) and particulate Mg, Li, Mn, U, V, As, Ba, Fe, Zn, Cd, Pb and Cu were measured using different techniques (HR-ICP-MS, ICP-AES and CCSA). Using in situ filtration as a baseline, we showed that ex situ filtration (on-board and on shore after freezing) resulted in an underestimation of the dissolved pool, which was counterbalanced by an overestimation of the particulate pool for almost all the elements studied. Analysis of major metallic elements in Chilean wines by atomic absorption spectroscopy Ciencia e Investigación Agraria

A set of 75 wine samples from seven of the major wine producing regions in Chile were analyzed with flame atomic absorption spectroscopy (AAS) to determine their content of major metallic elements (K, Mg, Ca, Na, Fe and Zn). The results obtained were further analyzed with principal component analysis (PCA) in an attempt to loosely discriminate these wines according to grape type and geographical origin. The metal concentrations measured were within normal ranges, according to previously published data from other wine producing countries. A adsorção de metais pesados no solo, tais como o cádmio e o cobre, é controlada por reações químicas e por processos físicos e biológicos. A retenção de metais pesados no solo pode diminuir a biodisponibilidade desses elementos no sistema solo-solução. Avaliou-se a adsorção de cádmio e de cobre em amostras de horizontes superficiais e subsuperficiais de oito solos, sendo cinco Latossolos, dois Nitossolos e um Argissolo. Avaliação da distribuição de cobre associado a compostos de diferentes massas moleculares na polpa de açaí Acta Amazonica

Neste estudo, cromatografia de exclusão por tamanho (SEC) com detecção por UV e detecção off-line por espectrometria de absorção atômica em forno de grafite (GF AAS) foi usada para investigar a associação de cobre a espécies de alta massa molecular (HMW) e baixa massa molecular (LMW) presentes na polpa de açaí (Euterpe oleracea Mart.). A concentração total de cobre obtida nos digeridos da polpa de açaí foi 10,5 µg g-1. Cobre foi encontrado associado às frações de HMW e LMW, correspondentes às massas moleculares de 28,7, 2,6 e 0,43 kDa.

Cadmium toxicity on seed germination and seedling growth of wheat Triticum aestivum Biological Sciences

Cadmium toxicity in seed germination and seedling growth of wheat (Triticum aestivum, var. IAC-370) is assessed. The ecotoxicological effects of six experimental concentrations of cadmium (0.03; 0.06; 0.12; 0.6; 1.2; 2.4; 4.8 mM), and control were evaluated. All assays were performed in quadruplicates with 25 seeds per replication in Petri dishes. Responses for toxic effect comprised the variables germination percentage, index of velocity of germination (IVG), length of aerial section and root of the seedlings, green and dry mass of the seedlings. Results showed that T. aestivum seeds exposed to cadmium decreased their normal germination percentage as from 0.03 mM concentration, with a 31% reduction of germination percentage and a 20% decrease in IVG....

ABSTRACT This study evaluates the effects of exposure to manganese (Mn2+) for 96 hours at two different temperatures (24 and 27°C) on juveniles of Centropomus parallelus through the activities of glutathione S-transferase (GST) and catalase (CAT), micronuclei test (MN) and comet assay. The GST activity did not show any significant difference between the groups exposed to Mn2+ and the respective control groups; in contrast, a major increase in the CAT activity was observed at 27°C in the group exposed to Mn2+ compared to the control group. Extraction, partial characterization and susceptibility to Hg2+ of acid phosphatase from the microalgae Pseudokirchneriella subcapitata Scientia Agricola

Pseudokirchneriella subcapitata is a unicellular green algae widely distributed in freshwater and soils. Due to its cosmopolitan characteristic, its use is recommended by national and international protocols in ecotoxicity studies. The alteration of phosphatase activities by agriculture pollutants like heavy metals has been extensively used as a biomarker in risk assessment and biomonitoring. In this study, we compared the extraction of acid phosphatase from P. subcapitata by different procedures and we studied the stability, substrates specificity, kinetics and the effect of Hg2+ in the crude extract. Free-radical depolymerization with metallic catalysts has been investigated to depolymerize an exopolysaccharide (EPS), with a high molecular weight (> 10(6) g/Mol), produced by the bacterium Alteromonas macleodii subsp. fijiensis biovar deepsane. Three metals (copper, zinc and manganese) were used either alone or mixed. These metals led to the depolymerization of the high molecular weight bacterial polysaccharide with different efficiency. Interactions between Volatile Reduced Sulfur Compounds and Metals in the Seine Estuary (France) ArchiMer

Concentrations of volatile hydrogen sulfide (H2S), carbonyl sulfide (OCS), methane thiol (MeSH), dimethyl sulfide (DMS), and dimethyl disulfide (DMDS) in the Seine estuary (France) were investigated in spring 2005 using samples collected from the first meter beneath the surface. Levels of dissolved metals (Ag, Cd, Cu, Zn, Ni, Co, Pb), suspended particulate matter, and particular organic carbon were also assessed. Interspecific comparison of Cd bloaccumulation in European Pectinidae (Chlamys varia and Pecten maximus) ArchiMer

The uptake and loss kinetics of Cd were determined in two species of scallops from the European coasts, the variegated scallop Chlamys varia and the king scallop Pecten maximus, following exposures via seawater, phytoplankton and sediment using highly sensitive radiotracer techniques (Cd-109). Results indicate that, for seawater and dietary pathways, C. varia displays higher bioaccumulation capacities in terms of uptake rate from water and fraction absorbed from ingested food (assimilation efficiency) than Pecten maximus. Regarding sediment exposure, P. maximus displayed low steady-state Cd transfer factor (TFSS < 1); however, once incorporated, a very large part of Cd transferred from sediment (92%) was strongly retained within R maximus tissues....

Mycorrhizal effectiveness and manganese toxicity in soybean as affected by soil type and endophyte Scientia Agricola

Mycorrhizal plants may present Mn toxicity alleviation and this depends on the plant-endophyte-environment interaction. The effectiveness of three arbuscular mycorrhizal fungi (AMF) (Glomus macrocarpum, G. etunicatum, G. intraradices) and a control without AMF in two soils: Typic Rhodudalf with high Mn availability and a Typic Quartzipsamment, with low Mn availability, was evaluated in a time-course experiment at 3, 6, 9 and 12 weeks after soybean (Glycine max L.) seedling emergence. Cádmio foi determinado em mostos e vinhos de duas cultivares de uvas vim/eras (Cabernet Sauvignon e Saint Emilion). Amostras/oram retiradas periodicamente durante a fermentação alcoólica e também do bagaço da cv. Cabernet Sauvignon, das borras e das sementes resultantes das trasfegas e do produto final. As fermentações de cada cultivar sofreram adição de quantidades conhecidas de cádmio (5mg/l e 5mg/l) afim de verificar o seu comportamento em diferentes concentrações. Subcellular distribution of Ag, Cd, Co, Cu, Fe, Mn, Ph, and Zn in the digestive gland of the common cuttlefish Sepia officinalis ArchiMer

The subcellular fractionation of the digestive gland cells of the common cuttlefish Sepia officinalis was performed to investigate the distribution of metals between organelles and cytosol and the different cytosolic fractions separated by gel-filtration chromatography. Total metal concentrations vary over 3 orders of magnitude, ranging from dwt for Pb to dwt for Zn. With the exception of Cd, Co, and Cu, metals were mostly bound to the organelles.
